The Andy Reid-led side are 8-0 on the year, and the only unbeaten team left in the league.
That included a dramatic 30-24 overtime win over the Tampa Bay Buccaneers on Monday Night Football.
But during the game, Mahomes suffered a worrying ankle problem, which raised concerns.
He had thrown a touchdown pass late in the showdown, but landed awkwardly doing so.
The 29-year-old fell to the ground, wincing in visible pain and in discomfort.
The superstar QB was helped to the sidelines, where head coach Reid tried to take him out.
He wanted to put backup Caron Wentz in to give Mahomes time to access the situation.
But the MVP ace stayed in, driving his team on to another win, despite his ankle problems.
After the clash, Mahomes told reporters he feared the injury would swell up this week.
And that appears to be the case ahead of the Chiefs game against the Denver Broncos.
They take on their NFL rivals looking to stay undefeated at 1pm on Sunday afternoon.
Footage from practice on Friday showed Mahomes with strapping around his ankle.
It looked to be solidly taped up, suggesting the issue is still causing him problems.
Fans were worried about the situation, fearing he might not make the Broncos showdown.
One said: “Sit him. No need to make him play against the broncos.”
Another added: “Ankle watch is back!”
A third commented: “8-0. Why not give him a week of rest before Buffalo.”
And a fan suggested: “Why on earth would they not sit pat for a week.”
Reid told the media his QB had practiced all week and did not miss any reps.
Mahomes is having another sparkling season with the Chiefs, and is searching for his fourth Super Bowl ring.
